Urine metabolomics of giant panda (Ailuropoda melanoleuca)

摘要: 为探究大熊猫(Ailuropoda melanoleuca)尿液代谢物的差异性,采用液相色谱-质谱UPLC-Q-TOF/MS技术对20例大熊猫尿液代谢成分进行检测,并通过性别和年龄分组对其尿液代谢物进行差异分析。研究发现,在性别组间共鉴定出12种显著差异代谢物(P<0.05),其中,除芹菜素外其余11种代谢物的相对含量均在雄性尿液中更高。亚成年组与成年组间共鉴定出25种显著差异代谢物(P<0.05),仅核黄素和精氨酸的相对含量在亚成年个体尿液中更高;而亚成年与老年组间和成年组与老年组间分别共鉴定出10种和3种显著差异代谢物(P<0.05),其相对含量均在老年个体尿液中更高。这些显著性差异代谢物主要集中在氨基酸代谢、次生代谢物的生物合成及黄酮、黄酮醇和类黄酮的生物合成通路。对不同性别、年龄间大熊猫尿液代谢物进行差异性分析,筛选出的差异代谢物为大熊猫化学通讯机理和生理功能的研究提供理论基础。

Abstract: In order to explore the difference of metabolites in giant panda urine, 20 cases of giant panda urine metabolites were detected by liquid chromatography-mass spectrometry (UPLC-Q-TOF/MS), and differences in urine metabolites were analyzed according to gender and age. As results, 12 metabolites with significant difference (P<0.05)were identified between the gender group, and the relative expression of 11 metabolites except for apigenin was higher in male urine. A total of 25 significantly different metabolites (P<0.05) were identified between the sub-adult group and the adult group. Only the relative expression levels of riboflavin and arginine were higher in the urine of the sub-adult giant panda. Ten significantly different metabolites (P<0.05)were identified between the sub-adult group and the geratic group, and three significantly different metabolites were identified between the adult group and the geratic group. The relative expression of these substances was higher in the urine of elderly individuals. These metabolites with significant differences were mainly concentrated in the amino acid metabolism pathway, the biosynthesis pathway of secondary metabolites and the biosynthesis pathway of flavones, flavonols and flavonoids. The urine metabolites of giant pandas of different genders and ages were analyzed in this study.The differential metabolites obtained by screening can provide a theoretical basis for the study of chemical communication mechanism and physiological function of giant pandas.
